Acetil-CoA ( acetil coenzima A ) é uma molécula que participa de muitas reações bioquímicas no metabolismo de proteínas, carboidratos e lipídios . Sua principal função é entregar o grupo acetil ao ciclo do ácido cítrico (ciclo de Krebs) para ser oxidado para produção de energia. A coenzima A (CoASH ou CoA) consiste em um grupo β-mercaptoetilamina ligado à vitamina ácido pantotênico (B5) através de uma ligação amida e ADP 3'-fosforilado. O grupo acetil (indicado em azul no diagrama estrutural à direita) do acetil-CoA está ligado ao substituinte sulfidrila do grupo β-mercaptoetilamina. Esta ligação tioéster é uma ligação de "alta energia", que é particularmente reativa. A hidrólise da ligação tioéster é exergônica (-31,5 kJ/mol).

A CoA é acetilada a acetil-CoA pela quebra de carboidratos através da glicólise e pela quebra de ácidos graxos através da β-oxidação . Acetil-CoA então entra no ciclo do ácido cítrico, onde o grupo acetil é oxidado a dióxido de carbono e água, e a energia liberada é capturada na forma de 11 ATP e um GTP por grupo acetil. GTP é o equivalente de ATP e eles podem ser interconvertidos pela Nucleosídeo-difosfato quinase .

Konrad Bloch e Feodor Lynen receberam o Prêmio Nobel de Fisiologia e Medicina de 1964 por suas descobertas ligando o acetil-CoA e o metabolismo dos ácidos graxos. Fritz Lipmann ganhou o Prêmio Nobel em 1953 por sua descoberta do cofator coenzima A.
